Sex-specific responses in placental fatty acid oxidation, esterification and transfer capacity to maternal obesity

چکیده

Fatty acid metabolism and oxidation capacity in the placenta, which likely affects rate composition of lipid delivered to fetus remains poorly understood. Long chain polyunsaturated fatty acids, such as docosahexaenoic (DHA), are critical for fetal growth brain development. We determined impact maternal obesity on placental oxidation, esterification transport by measuring PhosphatidylCholine (PC) LysoPhosphatidylCholine (LPC) containing DHA mass spectrometry mother-placenta-baby triads well free carnitine acylcarnitine metabolites women with normal obese pre-pregnancy BMI. Placental protein expression enzymes involved beta-oxidation pathways, MFSD2a (lysophosphatidylcholine transporter) OCTN2 (carnitine syncytiotrophoblast microvillous (MVM) basal (BM) membranes were Western Blot. Maternal was associated decreased umbilical cord plasma LPC PC fractions male, but not female, fetuses. Basal membrane increased placenta males mothers. In female placentas, despite an MVM expression, a reduced MUFA-carnitine levels enzymes. speculate that lower DHA-PL circulation male offspring mothers, significant increase transporter LPC-DHA, may lead low needed development contributing neurological consequences more prevalent children. Female placentas have appear store FA through greater esterification, suggesting impaired function transfer

Mitochondrial fatty acid oxidation in obesity.

SIGNIFICANCE Current lifestyles with high-energy diets and little exercise are triggering an alarming growth in obesity. Excess of adiposity is leading to severe increases in associated pathologies, such as insulin resistance, type 2 diabetes, atherosclerosis, cancer, arthritis, asthma, and hypertension. This, together with the lack of efficient obesity drugs, is the driving force behind much r...

Effect of Maternal Obesity on Fetal Growth and Expression of Placental Fatty Acid Transporters

OBJECTIVE To explore the effects of maternal high-fat (HF) diet-induced obesity on fetal growth and the expression of placental nutrient transporters. METHODS Maternal obesity was established in rats by 8 weeks of pre-pregnancy fed HF diet, while rats in the control group were fed normal (CON) diet. Diet-induced obesity (DIO) rats and diet-induced obesity-resistant (DIR) rats were selected ac...

Free fatty acid oxidation in insulin resistance and obesity.

The growing worldwide epidemic of obesity and diabetes portends a significant increase in cardiovascular disease. Obesity is associated with insulin resistance, and there is growing evidence that these conditions independently increase the risk of heart failure. Changes in myocardial substrate utilization develop in obesity and insulin resistance, and are characterized by increased fatty acid o...
